Histogenesis and Histochemistry of Mucosa of Respiratory Tract in Developin Chick

Abstract
In order to study the histogenesis of mucous membrane of respiratory tract and histocheznistry of their mucosubstances, the respiratory tract were observed in the chick embryos at 4, 7, 10, 13, 16 and 19th days in incubation and those in the chickens at 1, 4, 7 and 10th days after hatching. The mucosubstances were stained with akian blue(pH 2.5¡­1.0), PAS, alcian blue(pH 2.5¡­1.0)-PAS and aldehyde fuchsin (pH 1. 7)-alcian blue(pH 2. 5).
The results obtained were as follows:
1. The laryngeal and tracheal primordia of chick embryo began to appear early in the 4th incubation day and bronchial primordia in the 7th incubation day as a cell mass, and the lumen of respiratory tract were canalize in the 10th incubation day. The lining epithelium of respiratory tract was composed of 2-4 layers of columnar cells in the 10^-16th incubation days and changed into a typical pseudostratified ciliated columnar epithelium in the 19th incubation day. The goblet cells of the respiratory tract began to appear in the 16th incubation day and the mucous glands in the 19th incubation day.
2. The mucosubstances in the goblet cells and intraepithelial mucous gland of respiratory tract in the chick embryo were mixed type of a approximately equal amount of neutral and acidic mucosubst¡©ance and the secreation and stainability of mucosubstance became progressively increased after hatching.
3. The acid mucosubstance in the respiratory mucosae in the chick embryos was composed of both sulfated and nonsulfated mucin with more of sulfated mucin in the early stage of development and more of nonsulfated mucin after hatching.
